我一直对膜蛋白的三维结构和功能解析很感兴趣，高中时听说人类破解了“基因天书”，我感到很震惊。现在才知道要想破解“人类天书”，路还很长，而且首先得明确什么是关键。不错，就是蛋白质。蛋白质是构成人体的重要组分，其作用非常广泛，体内绝大多数酶是蛋白质，仅这一点就可说明蛋白质的重要地位。当然，蛋白质还充当激素、结构物质等角色。蛋白质的一个不容忽视的作用是作为生物膜上的结构和功能物质。生物膜功能涉及物质运输、能量转换、信号转导等重要生命活动，其中无不涉及蛋白质的作用，蛋白质一方面保证了膜适当的流动性，还可以作为物质运输的载体或通道、胞外信号的受体、膜酶等等。由此，看来解析膜蛋白的三维结构和功能意义确实非同寻常。

膜蛋白的解析史上有两个重要事件：一个是米歇尔教授等在世界上第一个解析膜内在蛋白，他们以0.32nm的高分辨率给出了紫色细菌光和反应中心的三维结构，接着又对其功能进行了阐释。可以说其成果是膜蛋白解析领域的一个里程碑。另一个是麦金农对离子通道的解析和阿格雷对水通道的解析，他们都做出了非常杰出的成果，并且都获得了诺贝尔奖。

20xx年，您在Nature上发表了关于菠菜主要捕光复合物(LHC-II)的晶体结构的文章，1)在0.272nm分辨率测定了重要光合膜蛋白LHC-II的晶体结构， 发现了膜蛋白结晶的第三种类型； 2) 建立了包括膜蛋白、色素分子和脂分子在内的蛋白脂质体的完整的LHC-II结构模型， 提供了近三万个(29038)独立的精确的原子坐标； 3) 首次基于精确的结构数据对高等植物的光能吸收、传递和光保护等热点问题进行了探讨。这无疑是对植物光合作用研究的跨越性贡献，我非常敬佩您，更是您做研究的态度。也正是这一点，是我决定考取您的研究生。我会是实验室最勤奋的一位，是最耐得住寂寞的一位，最富创造性的一位。

当然，膜蛋白的解析并非易事。当前主要研究的大都是膜内在蛋白，其特点是整合在膜内，具有两亲特性，要想将其从膜上分离出来需要特殊的去垢剂及适当的条件才能不致失活或结构变化。当前解析膜蛋白的方法主要是X射线衍射法、NMR和电镜三维重建，其中X射线衍射法的最大特点是分辨率极高，可达0.1nm，这是由X射线的特性决定的。其高分辨率决定了X射线衍射法的重要地位。NMR则可以用来显示蛋白质构象变化的整个动态过程，但是对于非水溶性的蛋白却有局限性。当然，Klug发明的低温电镜三位重建技术也在一些方面发挥作用。总之，X射线衍射法还是最主要的，自X射线被伦琴发现到布拉格父子提出布拉格方程，然后建立X射线晶体学以来，X射线已经为人类做出了巨大的贡献，特别是在医学上的应用。当然，X射线也为数十人获得诺贝尔奖创造了条件。

当前，摆在众多人面前的难题便是如何获得能够用于X射线衍射的蛋白质晶体(一般是单晶)。单晶要求高纯度、无孪晶、方向性好等。

米歇尔教授在完成紫色细菌光合反应中心的解析之后，专门出了一本书讲蛋白质结晶学。当然，蛋白质结晶过程极为复杂，受众多因素影响，蛋白质纯度、pH、温度、水活、载体等都很重要。有人曾在低重力条件下进行实验，效果很好。考虑到蛋白质晶体结构的维持大部分靠水分子间作用(含水30%-70%)，而膜蛋白的两亲性又使得其在水中难以形成稳定的规则排列的晶体，于是有人提出用另一种特殊溶液替代水。蛋白质结晶学现已成为一门坚难而有重大意义的学科，我认为如果谁能在此领域做出成绩，其意义远大于10个乃至100个膜蛋白的解析。 以下是我的简介：

2)黄豆脲酶的提取及动力学参数测定等，收获是掌握了脲酶的提取及纯化、分子量测定、最适温度及pH测定、抑制剂研究方法等；

3)太白参对苯丙芘导致的小鼠细胞DNA损伤的抑制作用，收获是掌握了PBS、细胞裂解液等的配制，最重要的是初步掌握了单细胞凝胶电泳法分析损伤DNA，另外还有一些氧化还原酶活性的测定；

4)参与关于肠道菌群平衡的体外模拟研究方案的设计及化学基因组学中小分子钓靶蛋白的实验方案设计。

5)参与苯并芘对乳腺癌细胞中甲基化影响的研究方案的设计，并即将投入研究。
